Hippocampal long-term depression mediates spatial reversal learning in the Morris water maze.

چکیده
Synaptic plasticity at hippocampal excitatory synapses has been proposed as the cellular mechanism underlying spatial learning and memory. However, most previous studies have focused on the role of long-term potentiation (LTP) in learning and memory, and much less is known about the role of long-term depression (LTD). Here, we report that hippocampal-dependent spatial learning in the Morris water maze facilitated hippocampal CA1 LTD induction in vivo. The LTD can be blocked by systemic application of the selective GluN2B antagonist Ro25-6981 (6 mg/kg, i.p.) or a synthetic peptide Tat-GluA2(3Y) (3 μmol/kg, i.p.) that interferes with the endocytosis of AMPA receptors. In addition, systemic or intrahippocampal administration of these two mechanistically and structurally distinct inhibitors impaired spatial reversal learning of a novel target location, when the hidden platform was moved to the quadrant opposite the initial target location. Notably, acute elevated-platform stress, which facilitates hippocampal LTD induction, enhanced both acquisition and retrieval of spatial reversal memory. The present study demonstrates that reversal learning is impaired by blocking hippocampal LTD, and enhanced by facilitating hippocampal LTD, suggesting that hippocampal LTD may be necessary and sufficient to mediate new information processing. Introduction: There are evidences showing the role of nitric oxide in the opiate reward properties. The role of nitric oxide signaling pathway as an intracellular mechanism on augmentation of long term potentiation in hippocampal CA1 area of rats is also confirmed. It has been also reported that oral morphine dependence facilitates formation of spatial learning and memory via activation of N...

It has been shown that brain glucose metabolism impairment, obesity, and diabetes couldlead to cognitive decline and Alzheimer’s disease (AD) pathogenesis. Kisspeptin (KP) a G-proteincoupled receptor neuropeptide, has been suggested as a link between energy balance andreproduction. Some studies have shown that the attenuation of KP signaling decreases metabolismand energ...

متن کاملSelective loss of hippocampal granule cells following adrenalectomy: implications for spatial memory.
We examined the effects of long-term adrenalectomy (ADX) on hippocampal anatomy and behavioral learning in two spatial memory tasks. We assessed damage throughout the hippocampus by stereological analysis of the dentate gyrus and Ammon's horn. Rats were ADX or sham operated, and then tested in the Morris water maze 12 weeks after surgery, followed by testing on an eight-arm, alternating-baited ...
